Profile Compound Selectivity Against 16 CDKs in Live Cells
The NanoBRET® Target Engagement (TE) CDK Selectivity Systems quantitatively measure intracellular compound affinity and occupancy against two panels of 8 CDKs each, allowing you to measure up to 16 CDKs in a single experiment. Each vector panel contains 8 individual tubes of CDK-NanoLuc® fusion vectors that are premixed with either a cyclin expression vector or carrier DNA for optimal transfections.
The CDK Selectivity Systems rely on the NanoBRET® TE technology, a bioluminescence resonance energy transfer (BRET) method that uses the small, extremely bright NanoLuc® luciferase as the energy donor and a cell-permeable NanoBRET® TE Tracer as the energy acceptor. The CDK Selectivity Systems use an addition-only, multiwell plate format with a single NanoBRET® TE Kinase Assay.
How the NanoBRET® TE CDK Selectivity Systems Work
A CDK-NanoLuc® fusion vector is co-transfected with a cyclin expression vector into cells. Within the cells, the co-expression drives association of the CDK and cyclin, creating a biased readout specific for the CDK partner protein pair. BRET is achieved by luminescent energy transfer from NanoLuc® luciferase to the cell-permeable NanoBRET® TE Tracer that is bound to the CDK-NanoLuc® fusion protein. When the test compound is added, it competes with the tracer. This results in a dose-dependent decrease in NanoBRET® signal, allowing intracellular affinity of the test compound to be quantitated.

Ready-to-Use Panels Simplify Cellular CDK Profiling
The NanoBRET® TE CDK Selectivity Systems simplify routine selectivity screening with ready-to-use CDK Vector Panels. Each panel contains 8 individual tubes of CDK-NanoLuc® fusion vectors that are premixed with either a cyclin expression vector or carrier DNA for optimal transfection (see tables). Just thaw, dilute and transfect these consumable vector mixes. Panel A contains the highly studied CDKs, and Panel B contains the understudied CDKs.
Turn-Key Cellular CDK Profiling System
The NanoBRET® TE CDK Selectivity Systems contain a NanoBRET® TE CDK Vector Panel (A or B) and a single NanoBRET® TE Intracellular Kinase Assay, K-10. The optimized assay protocol offers an easy, addition-only workflow. Begin by simply transfecting the vector panel into HEK293 cells in a multiwell plate. Following overnight incubation, add your test compounds to the assay plate and run the NanoBRET® TE Kinase Assay, K-10, using a single, optimized concentration of NanoBRET® Tracer K-10 for each CDK. A plate reader is used to measure BRET.
Get the Data You Need
Determine intracellular affinity and compound fractional occupancy with the NanoBRET® TE CDK Selectivity Assays.
The CDK4/6 inhibitors Palbociclib (Panel A and table) and Ribociclib (table) showed very selective engagement of CDK4/6, while Abemaciclib (Panel B and table) was CDK4/6-selective but also engaged CDK9 and the TAIRE family (table). Other inhibitors potently engaged CDK4/6 but also exhibited collateral engagement of other CDKs, including the TAIRE family. Affinities of >1µM are not reported in the table, although they are measurable as shown (Panel A).
Determine Compound Fractional Occupancy
The NanoBRET® TE CDK Selectivity Assays can quantitate the fractional occupancy of compound for each CDK.
Quantitative fractional occupancy measurements for Palbociclib and Abemaciclib. Measurements were obtained by including the appropriate experimental controls, so the BRET ratio can be converted to occupancy. Fractional occupancy enables profiling compound selectivity across the CDKs at a single compound concentration. Palbociclib and Abemaciclib are CDK4/6-selective inhibitors. The occupancy data for Palbociclib at 500nM and Abemaciclib at 500nM against CDK Vector Panel A and CDK Vector Panel B are shown (Mean and Standard Deviation were calculated from 3 independent experiments).
Featured Publication
In this study, the NanoBRET® TE method was used to profile a collection of 46 clinically advanced CDK inhibitors and tool molecules in live cells. The results confirmed the binding of advanced clinical compounds and revealed new drug-CDK interactions. These new findings offer the potential to repurpose drugs for lesser-studied CDK family members. This paper highlights the importance of determining drug selectivity across the CDK family in live cells. The NanoBRET® TE CDK Selectivity Systems expanded on this work, simplifying the workflow to use a single NanoBRET® TE Assay (K-10) and ready-to-use CDK Vector Panels.

User may use this product for research use only; however, no amplification or isolation of the vector components of this product and no transfer or commercial use of this product is allowed. (2a) use NanoBRET®-branded energy acceptors (e.g., NanoBRET® tracers, NanoBRET® dyes, BRET-optimized HaloTag® ligands) for all determinations of energy transfer activity; and
(2b) use NanoBRET®-branded LARs (e.g., NanoBRET® Nano-Glo® Substrate) or Intracellular TE Nano-Glo® Substrate/Inhibitor or Intracellular TE Nano-Glo® Vivazine/Inhibitor for all determinations of luminescent activity; or
(2c) contact Promega to obtain a license for use of the product for energy transfer assays using energy acceptors not manufactured by Promega. No license is needed if the energy transfer acceptor is a genetically encoded autofluorescent protein.
